Drug Description

Sinequan® (doxepin hydrochloride) belongs to the class of drugs called tricyclic antidepressants. It is a white crystalline solid solvable in water, lower alcohols and chloroform. It holds inactive ingredients such as Blue 1, Red 3, Red 40, Yellow 10, and other inert ingredients); magnesium stearate; starch and sodium lauryl sulfate.

Drug Indications

Generic Sinequan is a drug to treat depression and anxiety. It alleviates tension, elevate mood, improve sleep, increase energy, and usually alleviate the feelings of guilt, fear, and worry. It is successful in treating people whose depression and/or anxiety is psychological, related to alcoholism, or a result of psychotic depressive disorders (severe mental illness).

Drug Contraindications

Sinequan is contraindicated in patients having hypersensitivity to tricyclic antidepressants; glaucoma; concomitant use with MAOI; risk of urinary retention; and using it during acute recovery phase after MI.

Dosage Information

The usual starting daily dose is 75 mg. Dosage may consequently be raised or decreased at suitable intervals and based on individual response. The maximum dose range should range from 75 mg/day to 150 mg/day.

In severe cases, higher doses may be recommended which can be further increased to 300 mg/day if essential.

A lower dose of 25-50 mg/day can be considered in patients with very mild symptomatology or emotional symptoms accompanying organic disease.

A patient can take the total daily dosage on a divided or once-a-day dosage schedule. The maximum dose is 150 mg/day taken at bedtime if you want to take it once-a-day timetable.

Side Effects

Generic Sinequan may result in many unwanted side effects. If you experience any such reactions or change in intensity, tell your doctor immediately possible. Side effects that have been reported after using this drug include constipation, blurred vision, dizziness, dry mouth, drowsiness, itchy or scaly skin (pruritus), low blood pressure, light sensitivity, nausea, rash, trouble urinating, rapid or irregular heartbeat, and water retention.

Drug Interactions

Alcohol must be avoided while using Doxepin hydrochloride. This drug also should be avoided with drugs known as MAO inhibitors such as antidepressants phenelzine and tranylcypromine, tolazamide, cimetidine, quinidine, major tranquilizers, propafenone, flecainide, guanethidine, clonidine, carbamazepine and other antidepressants such as amitriptyline nefazodone.

If you are changing from fluoxetine, don't take this drug for at least 5 weeks after your last dose of fluoxetine.

Warning and Precaution

Sinequan should not be used for treatment of anyone less than 12 years old.

Major depression can worsen symptoms and/or emerge suicidal thinking or behavior in both adults and children, no matter if they are taking antidepressants.

Individuals who are using Sinequan and their caregivers should be monitored for any change in symptoms or any new symptoms that appear unexpectedly-especially agitation, hostility, anxiety, panic, extreme hyperactivity, restlessness, and suicidal thinking or behavior. Be especially cautious at the commencement of treatment or whenever you change a dose.

Sinequan may bring about drowsiness or less alert. Hence you should not drive or operate dangerous machinery or participate in any risky activity calling for full mental alertness.

Let your physician or dentist know that you are taking Sinequan before you have surgery or dental treatment.
